Toffoli was born in Scarborough, Ontario, to Rob and Mandy Toffoli; he has two older sisters Megan and Courtney. His father Rob was also involved in hockey and was the general manager of the Toronto Jr. Canadiens while Toffoli played with them. Growing up, Toffoli attended Birchmount Park Collegiate Institute in Toronto (into which Scarborough had amalgamated in 1998) and Hillcrest High School in Ottawa.

The Staples Center was electric, the kind of buzz that hummed in your chest before a playoff game. Toffoli skated out with the rest of the Kings, blue and gold, the crowd already roaring. He lined up at the top of the zone, stick on the ice, and waited. The puck dropped. A Kings defenseman fed him along the blue line, and he turned, shouldered into the opposing winger, and drove the net with that low, grinding speed that made the crowd lose their minds. The puck ricocheted off the crossbar, and he spun back, chest-to-chest with a defender, both of them sliding into the boards. The horn didn't matter. The roar did. He stood up, stuck the puck on his skate, and skated back to the bench with that quiet, satisfied nod. The Kings' fans were already chanting his name.

Toffoli was drafted seventh overall by the Ottawa 67's in the 2008 OHL draft. He was later drafted 47th overall by the Los Angeles Kings in the 2010 NHL entry draft. He signed a three-year entry-level contract with the Kings on April 19, 2011. He made his NHL debut on March 16, 2013, and scored his first NHL goal on March 18, 2013. He won the AHL's Dudley 'Red' Garrett Memorial Award for rookie of the year in 2012–13. Toffoli won the Stanley Cup with the Kings in 2014, assisting on Alec Martinez's Cup-winning goal. He was traded to the Vancouver Canucks on February 17, 2020, then signed with the Montreal Canadiens on October 12, 2020. He was traded to the Calgary Flames on February 14, 2022.
