Aegis Defenders blends 2D platforming with tower defense mechanics. Players control a team of up to two characters, each with a unique sub-ability and the capacity to build a distinct item. These items can be combined through "cross fusion" to create more powerful versions. Levels typically split into an exploration phase, involving platforming and puzzles, and a tower defense phase, where players defend a base—an important person or object—from waves of enemies. The story campaign includes 18 levels, plus two bonus levels featuring crossover characters from other Kickstarter games: Shovel Knight and Lunais from Timespinner.

The plot follows Ruinhunters Bart and his granddaughter Clu, who discover a powerful weapon called Aegis and set out to keep it from an evil empire. They are joined by Kobo the companion drone, Kaiim the monk, and Zula the bandit.

The game began as a student project by Bryce Kho and Lifu Lin for USC’s Intermediate Games class. After graduating, Kho founded the indie studio Guts Department to develop it further, funding the project through a 2014 Kickstarter campaign. The core development team consisted of three people: Bryce Kho as director, writer, artist, and designer; his sister Kristen Kho as engineer; and Max Palazzo as producer. Bryce's wife, Lai Xu, co-wrote the script, while Palazzo and Mike Effenberger assisted with level design and tuning. Music and sound effects came from Power Up Audio, with a soundtrack by Riley Koenig featuring 13 original tracks blending old-school synth with traditional Japanese RPG orchestral music. The game was demoed at PAX West 2017 Indie Megabooth, Tokyo Game Show 2017, Gamescom 2017, PlayStation Experience 2017, and SXSW 2018. It released on Windows and macOS via Steam on February 8, 2018, with PlayStation 4 and Nintendo Switch versions following on April 10, 2018.

Reception included praise from Destructoid's Chris Carter, who called it "a smooth, well-thought-out ride" with original art and charm. It was nominated for the 2018 SXSW Gamer's Voice Award and won "Best of the Mix" at E3 Week Mix 2017.
