The series centers on the conflict between the Maximals and Predacons over control of Angolmois Capsules. Big Convoy, a 'one-man army,' is tasked with teaching a group of young recruits to confront Magmatron's group. As the series progresses, both factions ultimately unite to combat the dark god Unicron and his sub-group, the Blentrons.

The Maximal team includes Big Convoy (woolly mammoth), Longrack (giraffe), Colada (cobra), Stampy (rabbit/scissors triple-changer), Break (penguin), Heinrad (tanuki), and Mach Kick (horse). The Predacon faction is led by Magmatron, who can split into three prehistoric creatures and recombine into Magmasaurus. Other Predacons include Guiledart, Saberback, Sling, Dead End, and Archadis.

Recurring characters include the navigational computers Navi and DNAVI, the cosmic entity Unicron, and the chimeric Blentrons (Drancron, Elphorpha, Ratorarta). Guest characters include Rockbuster, Randy, Sharp Edge, Bump, Survive, Killerpunch, Hydra, Crazybolt, Bazooka, Hardhead, and Lio Convoy, who returns from Beast Wars II to assist Big Convoy.
