D'Amigo played one season of college hockey at Rensselaer Polytechnic Institute, where he won the ECAC Rookie of the Year award and was named to the ECAC Rookie Team. He finished fifth nationally in points per game by a rookie and led RPI in game-winning goals. After signing an entry-level contract with the Maple Leafs in 2010, he was assigned to the Toronto Marlies of the AHL and later to the Kitchener Rangers of the OHL. He spent several seasons in the Maple Leafs organization, serving as an alternate captain for the Marlies during the 2013–14 season before making his NHL debut on December 5, 2013, in a 3–2 win over the Dallas Stars. He scored his first NHL goal on December 14, 2013, against the Chicago Blackhawks.

D'Amigo was traded to the Columbus Blue Jackets in July 2014 and later to the Buffalo Sabres in December 2014, but never played an NHL game for either team. He spent the 2014–15 and 2015–16 seasons in the AHL with the Springfield Falcons and Rochester Americans. Unable to attract an NHL contract, he moved to Finland's Liiga in 2016, playing for Ilves and later KalPa. He then played two seasons in Germany's DEL with ERC Ingolstadt, followed by a return to North America with the ECHL's Orlando Solar Bears and a brief stint with the AHL's Colorado Eagles. He later played for Düsseldorfer EG and Löwen Frankfurt in the DEL, and most recently played for HK 32 Liptovský Mikuláš in the Slovak Extraliga before retiring and becoming an assistant coach for the Elmira Aviators of the NAHL.

Jerry D'Amigo's career is notable for his early success at the collegiate and international levels, winning gold at the 2009 IIHF World U18 Championships and the 2010 World Junior Ice Hockey Championships, where he finished among the top scorers.
