WrestleQuest follows two protagonists, Muchacho Man and Brink Logan, as they aim to win championship titles in pro-wrestling. Muchacho Man, modeled after 'Macho Man' Randy Savage, is a newcomer unaware that wrestling is scripted, while Brink Logan, a nod to Bret 'The Hitman' Hart, wrestles for his father's company as a reliable 'jobber-to-the-stars'. Both catch the attention of Mr. L. F. Font, a stuffed elephant who owns the biggest wrestling organization PAW and seeks to control all wrestling through aggressive and illegal methods. Their paths cross as they fight to save pro-wrestling and reality itself.

Combat is turn-based and takes place before a live audience, with a Hype Meter reflecting audience engagement. Using the same moves repeatedly bores the audience, lowering the meter and strengthening enemies; varied moves and strategies increase the meter and unlock bonuses. Some enemies are defeated by reducing hit points to zero, while others require a pinning mini-game where players press buttons at the right time. Players can customize entrance routines with theme music, pyrotechnics, and effects, cut promos before boss fights, and choose from Hype Types such as Technician, Showman, and Powerhouse.
