Osako began his club career with Kashima Antlers, winning multiple domestic honors including the J.League Division 1 in 2009, Emperor's Cup in 2010, J.League Cup in 2011 and 2012, Japanese Super Cup in 2009 and 2010, and Suruga Bank Championship in 2012 and 2013. He moved to German side TSV 1860 Munich on 6 January 2014, scoring on his debut in a 1–1 draw against Fortuna Düsseldorf and finishing the season with 6 goals in 15 appearances. In June 2014, he transferred to 1. FC Köln on a three-year contract, later extending until summer 2020. He played for Köln in their relegation from the Bundesliga on 28 April 2018. On 16 May 2018, he joined Werder Bremen for the 2018–19 season, scoring on his debut in a 6–1 DFB-Pokal win against Wormatia Worms. In February 2019, he was voted the 2018 Japanese Footballer of the Year. He returned to Japan in August 2021, joining Vissel Kobe, with whom he won the J1 League in 2023 and 2024, the Emperor's Cup in 2024, and the J1 100 Year Vision League in 2026. Internationally, he was named in Japan's preliminary squad for the 2018 World Cup, scoring the second goal in a win against Colombia, making Japan the first Asian team to beat a South American team in World Cup history. He also played in the 2019 AFC Asian Cup, scoring four goals to help Japan reach the final, where they lost 3–1 to Qatar.
