Marleau was born in Swift Current, Saskatchewan, and grew up on his family farm near Aneroid. His parents recognized his talent when he played with older players at the 1995 Canada Games. He played junior hockey for the Seattle Thunderbirds of the WHL, where he was named captain in his second season and led the team to its first Western Conference Championship. He was drafted second overall by the San Jose Sharks in the 1997 NHL entry draft, behind Joe Thornton.

Marleau debuted immediately in the 1997–98 season and became the youngest player to ever play in the NHL in the modern draft era. He was named captain of the Sharks midway through the 2003–04 season. After the 2004–05 lockout, he signed a new contract and had a breakout season in 2005–06, recording career highs of 34 goals, 52 assists, and 86 points. He passed Owen Nolan's franchise marks for goals, assists, and points in 2006–07. Marleau was removed as captain in 2009 but later served as alternate captain.

After 19 seasons with San Jose, Marleau signed with the Toronto Maple Leafs in 2017, then returned to the Sharks in 2019, and was traded to the Pittsburgh Penguins before the 2020 Stanley Cup playoffs. He returned to San Jose for the 2020–21 season, his final season. His number 12 was retired by the Sharks on February 25, 2023. Internationally, he won gold medals with Canada at the 2010 and 2014 Winter Olympics.
