Mastantuono was born and raised in Azul, Buenos Aires Province, into a family of Italian Argentine background. He began football at age three for River de Azul, where his father coached. In 2017, he trialed with River Plate but declined a place to focus on tennis. He later joined River Plate in 2019, made his official debut in January 2024, and scored his first goal in February 2024, becoming River's youngest ever goalscorer. He moved to Real Madrid in August 2025 for €45 million, and was loaned to Fiorentina in August 2026.
