Castellini made his professional debut at U.S. Cremonese at age 17, appearing in Serie B and Serie C1. He signed with Torino FC in summer 2000, achieving promotion to Serie A in his first season with 33 appearances. From January 2004, he served a loan at Brescia Calcio, then moved to Spain's Real Betis on a five-year contract, where he struggled with injuries and never dislodged veteran Luis Fernández. He returned to Italy in summer 2006 with Parma F.C., becoming first choice and helping the team return to Serie A after one year. In 2009–10, he played as a wingback in a 3–5–2 or 5–3–2 formation. On 26 August 2010, he was loaned to AS Roma as cover for John Arne Riise. On 24 June 2011, he was loaned to UC Sampdoria, agreeing to a permanent deal on 9 July 2012. He played in all three major levels until retirement in 2016, representing A.S. Livorno Calcio, Cremonese, and Brescia.
