Hamhuis played major junior hockey with the Prince George Cougars of the Western Hockey League (WHL). After being selected 12th overall by the Nashville Predators in the 2001 NHL entry draft, he returned to the WHL for a fourth season, where he won the Bill Hunter Memorial Trophy and Four Broncos Memorial Trophy as the league's best defenceman and player of the year, respectively. He also received the CHL Defenceman of the Year Award. He made his professional debut with the Milwaukee Admirals of the American Hockey League (AHL) in 2002–03 and his NHL debut the following season with Nashville. Over five seasons with the Predators, he recorded a career-high 38 points in 2005–06 and led the team in ice time. As an unrestricted free agent in 2010, he signed a six-year contract with the Vancouver Canucks. In his first year with the club, he helped Vancouver reach the 2011 Stanley Cup Finals, where they lost to the Boston Bruins. Hamhuis also represented Canada internationally, winning a gold medal at the 2014 Winter Olympics and medals at World Junior Championships and World Championships.
