Vanek was born in Baden bei Wien, Austria, to a Slovak mother and a Czech father who emigrated from Czechoslovakia in 1982. He was raised in Zell am See and later in Graz, spending considerable time at ice rinks accompanying his father, a professional hockey player and coach. As a youth, he competed in the 1997 and 1998 Quebec International Pee-Wee Hockey Tournament with a team from Austria, earning the scoring title and most valuable player honours in 1997. In 1998, he moved to Lacombe, Alberta, to play North American bantam hockey, later recording 74 goals and 36 assists for the Lacoka Wolves in the North Central Midget AA hockey league.

After playing junior hockey for the Sioux Falls Stampede of the USHL, Vanek joined the University of Minnesota Golden Gophers, leading the team in goals, assists, and points in the 2002–03 season and winning the 2003 NCAA National Championship. He was named MVP of the Frozen Four tournament and Minnesota's team MVP, becoming the first freshman to receive the honor. He spent the 2004–05 season with the Sabres' AHL affiliate, the Rochester Americans, leading the team in goals and power play goals. In the NHL, he recorded his best statistical season in 2006–07 with 43 goals and 41 assists, earning the NHL Plus-Minus Award. He was named to his first NHL All-Star Game in 2009 and made NHL history by becoming only the second player to score four consecutive natural hat-tricks.
