Masiello graduated from Juventus' youth system and made his Serie A debut for Juve in a home defeat to Inter on 15 May 2005. He joined Juventus from Lucchese in a co-ownership deal in 2003, and in January 2004 turned to a co-ownership deal for €300,000. In June 2004 Juventus acquired him outright. He was on loan to Avellino of Serie B in the 2005-06 season, playing 41 times.

Avellino lost the Serie B relegation playoffs to AlbinoLeffe 4–3 on aggregate. In the wake of the 2006 Italian football scandal, Masiello was joint-owned by Siena and Juventus, valued at €350,000. After failing to make an appearance in the first half of the 2006 season, Juventus sold 50% of its ownership of Masiello to Genoa on 25 January 2007 for €1.25 million. He played 15 times and won promotion with Genoa.

Genoa eventually acquired Masiello outright for €1.08 million. After playing 4 times for Genoa, he went on loan to Bari. Masiello joined Bari in a co-ownership bid after the 2007–08 season for €1.5 million. He was part of the team that won Serie B in 2009, and Bari bought him outright for another €800,000.

He moved to centre-back from right-back after Andrea Ranocchia's injury. On 15 May 2011, in the derby between Bari and Lecce, he deflected a shot into his own goal, ensuring Lecce's victory and survival. He later admitted he was offered at least €50,000 to turn the ball into his own net. On 25 July 2011, Bari sold Masiello to Atalanta for €2.5 million cash plus Marino Defendi (tagged for €1 million).

Bari retained 50% registration rights. He scored his first goal for Atalanta in a 2–2 draw with Fiorentina. After the exposure of the betting scandal, Masiello was released by the club, but returned shortly after. On 29 January 2020, Masiello signed a deal with Genoa. On 5 September 2022, he joined Südtirol on a one-season contract.
