Murzyn began his junior career in the Alberta Junior Hockey League with the Calgary Spurs before moving to the Western Hockey League's Calgary Wranglers. After a 31-point rookie season, he exploded for 32 goals and 92 points in his second year, leading all team defencemen in scoring and earning a spot on the WHL West First All-Star Team. He was the second-ranked prospect for the 1985 NHL entry draft and was selected fifth overall by the Hartford Whalers.

In the NHL, Murzyn transitioned from a prolific junior scorer to a stay-at-home defenceman known for his strength in front of the net and in the corners. After two and a half seasons with Hartford, he was traded to his hometown Calgary Flames in January 1988. His first full season with Calgary culminated in a Stanley Cup championship in 1989. He later recorded a hat trick in a 12–2 win over the Toronto Maple Leafs in February 1990.

Murzyn was traded to the Vancouver Canucks at the 1991 trade deadline. In Vancouver, he partnered with offensive defenceman Jyrki Lumme as the team's top defensive pairing, a duo that became the league's longest-enduring until Murzyn's retirement. He won the Babe Pratt Trophy as the Canucks' best defenceman in 1993, posting a career-high +34 plus-minus rating. During the 1994 playoffs, the Canucks reached the Stanley Cup Final, but Murzyn missed the series due to two separate injuries. Knee problems hampered the remainder of his career, and he retired in 1999 after splitting his final season between the Canucks and their AHL affiliate.
