Holloway was born in Honolulu, Hawaii, and raised in Waiʻanae. He is of Native Hawaiian and Samoan descent. He started training in kickboxing in 2007 at age 15 and graduated from Waiʻanae High School in 2010. He holds UFC records for most significant strikes (3,681) and total strikes (3,980). He also owns multiple UFC featherweight records, including most wins (20), longest winning streak (13), most finishes (11), and most knockout victories (9).

Holloway made his UFC debut at UFC 143 against Dustin Poirier, losing via submission. He later went on a win streak, defeated Anthony Pettis for the interim featherweight championship, and unified the title by defeating José Aldo at UFC 212. He has fought at lightweight for an interim title against Dustin Poirier and was scheduled for a lightweight title fight against Khabib Nurmagomedov but was pulled due to weight cut issues.
