The band takes its name from a drinking club consisting mainly of Cooper, John Lennon, Keith Moon, Micky Dolenz, and Harry Nilsson. The mission of the original Hollywood Vampires was to drink until no one could stand up, during the height of Cooper's drinking in the 1970s. Cooper discusses the Rainbow Bar incarnation of the Hollywood Vampires in the 1991 video biography Prime Cuts.

The Hollywood Vampires debuted live at the Roxy Theatre in West Hollywood on September 16 and 17, 2015, with guest performers including Tom Morello, Geezer Butler, Perry Farrell, Zak Starkey, Kesha, and Marilyn Manson. The group performed at Brazil's Rock in Rio festival on September 24, 2015, and at the Grammy Award ceremony in February 2016 as a tribute to Lemmy. Their first concert tour began in May 2016, and they made late-night television appearances on Jimmy Kimmel Live! in 2019. In 2023, the band toured Europe and the United States, with a final show at Bethel Woods Center for the Arts on July 30, 2023, the site of the 1969 Woodstock Festival.
