Hypoplectrus puella was first formally described in 1828 by French anatomist and naturalist Georges Cuvier, who named it Plectropoma puella based on a type collected at Martinique. In 2012, two new species of 'barred hamlet' were described: Hypoplectrus ecosur from the Yucatan and H. floridae from the eastern Gulf of Mexico.
