The story picks up after *Lobo*, a 2008 fantasy series that also featured Angel Locsin alongside Piolo Pascual. That earlier show dealt solely with werewolves, with no vampires involved. In *Imortal*, Locsin plays Lia, a character unrelated by blood to her *Lobo* character Lyka.
The plot begins in 1572, when vampires—beings that feed on human blood—arrive in the Philippines and spread terror. Unbeknownst to them, the islands are already home to the Taong Lobos, a clan of shapeshifters who can transform into powerful wolves. These werewolves act as protectors of humans, and together with people, they form an army to fight the vampires. The blood drinkers are overwhelmed, and most are destroyed; the survivors go into hiding. Peace returns, and the memory of vampires fades. During this time, the Taong Lobos face their own internal crisis, which sets the stage for the events of *Lobo*.
But the vampires have not vanished for good. They have been lying low to rebuild their strength, holding onto a secret prophecy. This prophecy foretells an ultimate battle between vampires and Taong Lobos that will decide the fate of both bloodlines. The conflict will be led by a powerful vampire and a powerful werewolf, each destined to be the other's downfall. The vampires believe they have found their prophesied champion and are preparing for war. The Taong Lobos, however, remain unaware of the prophecy and have grown careless.
Into this brewing conflict come two individuals: Mateo (John Lloyd Cruz) and Lia (Angel Locsin). Drawn to each other, they unknowingly awaken dormant powers within themselves—powers that will ultimately drive them apart.
