Suzette Doctolero was raised in Calabanga, Camarines Sur, and is the granddaughter of Buenviges Narvadez, a Filipino comedienne and actress from the 1930s. She attended high school at José Rizal College (later José Rizal University) and studied at the Polytechnic University of the Philippines (PUP), where she took courses in accounting, clinical psychology, and finally a bachelor of arts in Filipino. While at PUP, she joined theater groups Dulaang Kalayaan and Dulaang Bonificacio, portraying Gregoria de Jesus in one play.

At age 19 in 1988, her adviser Angie Ferro hired her as a production assistant for Balintataw TV. She later wrote romantic novels, and with help from Lualhati Bautista, her work Ako si Alex, Babae was published by Anvil Publishing. In the 1990s, she sold her works to various publishers and contributed to Viva Television shows. She became a screenwriter for GMA Network, working on Kirara, Ano Ang Kulay ng Pag-ibig?, and Sana Ay Ikaw Na Nga. Encantadia was her first project as head writer and her first in the telefantasya genre.

Doctolero also wrote for Daisy Siete and adapted Lupin, Joaquin Bordado, Totoy Bato, Gagambino, and Panday Kids, though most of these adaptations did not achieve high ratings. In 2016, she served as creative consultant for Alyas Robin Hood, which drew comparisons to Arrow; she defended the show, stating it was based on the public-domain legend of Robin Hood. In 2017, she was creative consultant for Destined to be Yours, and her tweets about its early ratings prompted an official statement from GMA Network distancing itself from her personal opinions.
