Born in New York City to a Jewish family, Landau grew up in Brooklyn and Queens before moving to Lexington, Massachusetts at age 12. He attended Lexington High School and Brandeis University, earning a degree in history with honors. Aligning himself with the underground culture of late-1960s Boston, he wrote for the music magazine Crawdaddy, championing straightforward rock and roll while criticizing the San Francisco scene. As a critic for Rolling Stone, he wrote about Jimi Hendrix, Cream, Aretha Franklin, Sam and Dave, and Otis Redding.

Landau's 1974 article in The Real Paper, in which he wrote 'I saw rock and roll's future and its name is Bruce Springsteen!,' is credited with fostering Springsteen's popularity. He was subsequently hired by Springsteen and co-produced studio records from 1975's Born to Run through 1992's Human Touch and Lucky Town. Landau has occasionally joined Springsteen onstage to play guitar. In 2024, a film based on the making of Springsteen's 1982 album Nebraska was announced, with Jeremy Strong playing Landau; the film was released in 2025.

Landau has managed or produced other artists including MC5, Livingston Taylor, Jackson Browne, Natalie Merchant, Alejandro Escovedo, Train, and Shania Twain. He has written liner notes for collections by Aretha Franklin, Otis Redding, and Wilson Pickett. He received the Rock and Roll Hall of Fame's Ahmet Ertegun Award for Lifetime Achievement in 2020.
