Gasperini entered the Juventus youth system at age 9, winning an Allievi Nazionali championship and being part of the Primavera squad that placed runner-up in 1976. He played a handful of Coppa Italia matches with the Juventus first team, was loaned to Reggiana, then sold to Palermo in 1978. He stayed five seasons at Palermo in Serie B, reaching a Coppa Italia final in 1979. After spells with Cavese and Pistoiese, he moved to Pescara, making his Serie A debut in a 2–1 win over Pisa in which he scored. He later played for Salernitana and retired in 1993 after two seasons with Vis Pesaro.

Gasperini began his managerial career at Juventus's youth system in 1994, coaching the Giovanissimi, Allievi, and Primavera. In 2003 he became head coach of Crotone, guiding them to promotion to Serie B. He then managed Genoa, leading them to Serie A promotion and a fifth-place finish in 2008–09, securing a Europa League spot. After a poor start in 2010–11, he was dismissed. He managed Inter Milan in 2011 but was sacked after five winless games. He had two brief spells at Palermo in 2012–13, then returned to Genoa in 2013. In 2016 he became Atalanta manager, leading them to five Champions League qualifications and winning the Europa League in 2024. He left Atalanta in June 2025 and was appointed Roma manager.
